It activates both ERK1/2 and PKCα signaling pathways, leading to phosphorylation of Bcl-2 at Ser70 and c-Myc at Thr58 and Ser62, thereby promoting cell survival and proliferation in human lung cancer cells. Due to its strong tumorigenic potential, NNK is widely used to induce lung cancer in experimental mouse models for studying carcinogenesis and evaluating anticancer therapeutics.
