It acts as a multireceptor antagonist, with binding affinities (Kd values) of 102 nM, 180 nM, 180 nM, and 150 nM for the 5-HT2A, dopamine D2, α1-adrenergic, and α2-adrenergic receptors, respectively. With lower binding affinity for histamine H1 and various serotonin receptors, Melperone also exhibits inhibitory effects on CYP2D6.
