JMI-105 is a potent inhibitor of PfFP-2, a protease of Plasmodium falciparum critical for malaria pathogenesis. It demonstrates inhibitory activity against both chloroquine-sensitive (3D7; IC50=8.8 µM) and chloroquine-resistant (RKL-9; IC50=14.3 µM) strains of P. falciparum. In preclinical studies, JMI-105 significantly reduced parasitemia and extended survival in a murine model infected with P. berghei ANKA, indicating its potential as an effective anti-malarial therapeutic agent.
