PL265 is a dual enkephalinase inhibitor that acts as a prodrug of PL254. It effectively inhibits both neutral endopeptidase (neprilysin) and aminopeptidase N, leading to increased local concentrations of enkephalins like Met-Enkephalin and Leu-Enkephalin. This action subsequently activates μ and δ opioid receptors, resulting in a potent analgesic effect. Additionally, PL265's inhibition of leukotriene A4 hydrolase (LTA4H) may further enhance its anti-inflammatory properties by reducing the production of leukotriene B4 (LTB4).
