Serpin B9-IN-1 (BTCA) is a selective inhibitor targeting Serpin B9 (SB9), a natural inhibitor of granzyme B (GrB) that is implicated in promoting lung cancer cell metastasis to the bone marrow. Elevated levels of SB9 in cancer cells enhance their proliferation and metastatic potential through interaction with GrB in an immune cell-dependent manner. Inhibition of SB9 by Serpin B9-IN-1 has been shown to significantly impair immunotherapy responses for lung cancer with bone metastases in the caudal artery (CA) mouse model, making it a valuable reagent for cancer research focused on the tumor microenvironment and immune evasion.
