ASP-8731 is a potent BACH1 inhibitor that enhances NRF2-mediated gene transcription, thereby activating antioxidant and anti-inflammatory pathways. This compound significantly upregulates the expression of key genes such as HMOX1 and FTH1, and increases fetal hemoglobin (HbF) levels, promoting F-cell production in hydroxyurea-unresponsive cells. Additionally, ASP-8731 mitigates inflammatory responses by downregulating VCAM1, ICAM-1, and NF-κB (p65) phosphorylation.
