E1/E2/E3 Enzyme Inhibitor
1-Benzyl-I3C functions as a potent inhibitor of the NEDD4-1 enzyme, exhibiting significant anticancer properties. With an IC50 of 12.3 μM, it markedly surpasses the efficacy of its precursor, I3C. The compound effectively inhibits the proliferation of human melanoma cells, correlating with its ability to inhibit NEDD4-1's ubiquitination activity. Binding studies, including in vitro ubiquitination assays and thermal stability analyses, confirm that 1-Benzyl-I3C interacts with the catalytic HECT domain of NEDD4-1, providing insights into its mechanism of action.
