AGPAT4 Inhibitor
AGPAT4-IN-1 is a covalent inhibitor of AGPAT4 that targets Cys228, resulting in an IC50 of 795 nM. This compound effectively inhibits acyltransferase activity, disrupting the conversion of lysophosphatidic acid (LPA) to phosphatidic acid (PA) and the downstream mTOR/S6K signaling pathways. AGPAT4-IN-1 enhances the sensitivity of hepatocellular carcinoma (HCC) tumors to Sorafenib and promotes apoptosis, demonstrating a synergistic effect. Additionally, this reagent exhibits antitumor activity by reducing tumorigenicity and stemness in HCC xenograft models.
