Akuammine

Akuammine is an indole alkaloid primarily known for its analgesic activity, obtained from Picralima nitida. It selectively binds to the μ- and κ-opioid receptors, demonstrating affinities of 0.3 μM and 1.68 μM, respectively, while showing less affinity for the δ-opioid receptor (10.4 μM). Akuammine effectively inhibits forskolin-induced cAMP production in HEK293 cells expressing these opioid receptors, with IC50 values of 2.6 μM for μ-opioid and 0.073 μM for κ-opioid receptors. In vivo studies reveal that akuammine enhances pain thresholds in mice, as evidenced by increased latency in tail-flick and hot plate tests following administration at a dose of 60 mg/kg.
