Synthetic Lethal Lead

AL-GDa62 is a synthetic lethal lead that targets apoptosis pathways in CDH1-deficient cells. This compound demonstrates potent activity with an EC50 of 3.2 μM in wild type MCF10A mammary epithelial cells and 2 μM in CDH1-/- mutant cells. AL-GDa62 specifically inhibits TCOF1, ARPC5, and UBC9, effectively suppressing SUMOylation at low micromolar concentrations. Its unique mechanism of action makes it a valuable tool for research into cancer therapies targeting E-cadherin loss.
