Americanin A

Anticancer Agent

Americanin A is a neolignan that exhibits potent anticancer activity by activating the ATM and ATR pathways, which in turn initiate critical signaling cascades involving Chk1, Chk2, and the tumor suppressor p53. It selectively targets Skp2 for degradation, leading to the stabilization of the cyclin-dependent kinase inhibitor p27, and suppresses the activity of Cyclin B1 and cdc2 to inhibit cell cycle progression into mitosis. Additionally, Americanin A induces apoptosis through the production of excessive reactive oxygen species (ROS) and demonstrates efficacy against colorectal cancer.
