Amorphigenin

c-Fos/NFATc1 Inhibitor

Amorphigenin is a c-Fos and NFATc1 inhibitor that plays a critical role in the regulation of osteoclast differentiation. This compound acts by suppressing the expression of key factors in activated T cells, thereby mitigating inflammation-related bone loss. Additionally, Amorphigenin promotes the degradation of melanosome proteins through an AMPK-dependent autophagy pathway, independent of mTOR signaling. Its protective effects on bone mass and reduction of bone erosion in murine models make it valuable for investigating inflammatory bone diseases, postmenopausal osteoporosis, and disorders of skin pigmentation.
