TREM2 Agonist
As48 is a selective TREM2 agonist that demonstrates a KD value of 12.48 μM in TRIC binding assays. By binding to the TREM2 cleavage region, As48 enhances SYK phosphorylation, increases microglial phagocytosis, and activates downstream calcium signaling in TREM2-expressing cells. Additionally, As48 inhibits TREM2 ectodomain shedding while not interfering with ADAM10/17 protease activities. This compound serves as a valuable tool for investigating mechanisms associated with Alzheimer’s disease.
