Axl-IN-21

AXL Inhibitor

Axl-IN-21 is a selective, orally active inhibitor targeting the AXL receptor tyrosine kinase with a Kd of 2.7 nM and an IC50 of 4.0 nM. It demonstrates significant kinase selectivity and exhibits potent activity against various cancer-related multikinases, including Mer (Kd = 1.4 nM) and DDR1 (IC50 = 22.2 nM). Axl-IN-21 effectively blocks GAS6-induced AXL/STAT3/ABCG1 signaling, which enhances chemosensitivity and mitigates drug efflux in gastric cancer. Furthermore, this compound inhibits TGF-β1-mediated epithelial-mesenchymal transition, migration, and invasion in MDA-MB-231 cells, while showing minimal cytotoxicity to non-cancerous cells, making it a promising candidate for research in triple-negative breast cancer and gastric cancer.
