C14-490 is an ionizable cationic lipid with a pKa of 5.94, primarily utilized in the formulation of lipid nanoparticles (LNPs). These LNPs serve as a delivery system for encapsulating SpCas9 mRNA and TTR sgRNA, facilitating in utero gene editing specifically in hematopoietic stem cells (HSCs). C14-490-based LNPs, optimized through the B5 formulation and surface conjugation with CD45 antibody F(ab’)2 fragments, are part of the systematically optimized Targeted Editing Machinery LNPs (STEM LNPs), enhancing the precision of gene editing techniques.
