Chromomycin A3
Chemical Sensitizer
Chromomycin A3 is a chemical sensitizer that selectively binds to GC-rich DNA sequences, targeting the DNA minor groove upon dimerization with Mg2+. This compound inhibits DNA replication and transcription, impeding the binding of the Sp1 transcription factor to gene promoters, leading to the downregulation of anti-apoptotic proteins like FLIP, Mcl-1, and XIAP. Chromomycin A3 induces S-phase cycle arrest and engages caspase-dependent apoptosis in tumor cells. Additionally, it can mitigate oxidative stress from glutathione depletion and neuronal apoptosis prompted by Camptothecin. This agent is applicable in fundamental research involving malignant tumors, including cholangiocarcinoma, and serves as a potential probe for GC-rich regions.
