MTHFD2 Inhibitor
DS44960156 is a selective inhibitor of methylenetetrahydrofolate dehydrogenase 2 (MTHFD2), exhibiting an IC50 of 1.6 μM and a Ki of 1.23 μM. This compound binds to the active site of MTHFD2, disrupting redox homeostasis and blocking serine-mediated one-carbon metabolism, leading to an enhanced NAD+/NADH ratio and increased levels of reactive oxygen species (ROS). DS44960156 effectively inhibits glioma cell proliferation and sensitizes cells to glutamine starvation-induced cytotoxicity. Additionally, it has been characterized as non-mutagenic and non-carcinogenic, making it a valuable research tool for studying glioblastoma multiforme and related malignancies.
