GDC-0134

DLK Modulator

GDC-0134 is a selective modulator of dual leucine zipper kinase (DLK) that effectively crosses the blood-brain barrier. By inhibiting DLK's kinase activity, GDC-0134 disrupts the activation of the downstream JNK signaling pathway, thereby preventing DLK-dependent retrograde signal transduction associated with axon-to-soma degeneration and providing neuroprotective effects. This compound has shown promise in reducing TDP-43 protein aggregation and mitigating neuromuscular junction denervation in motor neurons, making it a valuable tool for investigating amyotrophic lateral sclerosis (ALS), Alzheimer's disease, and other neurodegenerative disorders linked to DLK activity.
