GL64

ADGRD1 Agonist

GL64 is a selective agonist of ADGRD1, exhibiting an EC50 of 3.98 μM. It has some low selectivity towards isoforms such as ADGRD2 and various CELSR and ADGRG proteins. By mimicking the satchel sequence, GL64 activates ADGRD1, leading to the regulation of osteoclast maturation through the cAMP-PKA-NFATC1 signaling pathway. This compound demonstrates the ability to inhibit osteoclastogenesis and prevent bone loss, making it valuable for researching osteoclast-related diseases.
