HPA-12

CERT Inhibitor

HPA-12 is a ceramide transfer protein (CERT) inhibitor that effectively permeates the blood-brain barrier. This small-molecule compound disrupts the transport of ceramide from the endoplasmic reticulum to the Golgi apparatus by binding to the START domain of CERT, resulting in increased intracellular ceramide levels and reduced sphingomyelin synthesis. HPA-12 induces endoplasmic reticulum stress through the GRP78/ATF6/CHOP signaling pathway and promotes mitochondrial autophagy, which can suppress cell proliferation and trigger apoptosis. In preclinical studies, HPA-12 demonstrates substantial efficacy in reducing leukemia burden and prolonging survival in mouse models of acute myeloid leukemia, making it a valuable tool for investigating lipid metabolism in conditions such as acute myeloid leukemia and Alzheimer's disease.
