IDB-001

PTC Inhibitor

IDB-001 is a potent inhibitor of the human ribosomal peptidyl transferase center (PTC), disrupting translational elongation through conformational changes induced by specific interactions with Asp/Glu residues in nascent polypeptides. This compound preferentially stalls ribosomes at sites with acidic peptide motifs, leading to cancer cell proliferation inhibition and activation of the integrated stress response via eIF2α phosphorylation at elevated concentrations. Furthermore, IDB-001 can induce ribotoxic stress responses through JNK and p38 phosphorylation. It has been effectively utilized in mechanistic studies of triple-negative breast cancer.
