ABHD6/DAGL-α Inhibitor
LEI-106 is a dual inhibitor of both ABHD6 and DAGL-α, with a Ki of 0.8 μM for ABHD6 and an IC50 of 18 nM, along with a Ki of 0.7 μM for DAGL-α. This compound effectively inhibits the synthesis of 2-arachidonoylglycerol, leading to a reduction in endothelial cell-derived 2-arachidonoylglycerol levels without affecting cannabinoid and diacylglycerol concentrations. LEI-106 is relevant for investigations into diet-induced obesity and metabolic syndrome, providing valuable insights into lipid signaling pathways.
