Methyl cholate

TcdB Inhibitor

Methyl cholate is a bile acid analog that acts as a specific inhibitor of the TcdB toxin from Clostridioides difficile. It demonstrates a stronger selective inhibitory effect on TcdB compared to TcdA, facilitating conformational stabilization by binding to a unique site on TcdB. This action prevents the toxin from attaching to host receptors and undergoing self-processing, offering protection to human fibroblasts from TcdB-induced cytopathic effects. Methyl cholate also exhibits dose-dependent anti-hepatic fibrosis activity in cellular and zebrafish models by significantly reducing the expression of α-SMA and COL-I, making it valuable for research in Clostridioides difficile infections and hepatic fibrosis.
