METTL1-WDR4 Inhibitor
METTL1-WDR4-IN-1 is a selective competitive inhibitor of the METTL1-WDR4 methyltransferase complex, with an IC50 of 144 μM. This compound effectively inhibits m7G methyltransferase activity, resulting in decreased PKM2 protein expression and disruption of the METTL1/PKM2/H3K9la positive feedback loop. Additionally, METTL1-WDR4-IN-1 impairs PKM2 nuclear translocation and consequently inhibits CD155 transcriptional activation. It exhibits potential in reducing tumor cell proliferation, altering glycolytic metabolism, and enhancing immune responses by restoring NK cell and CD8+ T cell function. This reagent is valuable in the study of immunotherapy for colorectal cancer and is ideally combined with PKM2 inhibitors to improve anti-tumor efficacy.
