N-Propargylglycine

PRODH Inhibitor

N-Propargylglycine is a potent inhibitor of proline dehydrogenase (PRODH). This inhibitor covalently modifies enzyme-bound FAD and active site lysine, leading to structural alterations, protein decay, and irreversible inhibition of proline and 4-hydroxyproline catabolism. N-Propargylglycine is involved in mitochondrial stress responses and enhances mitochondrial chaperone levels, thereby promoting proteostasis. It has demonstrated efficacy in models of Huntington's disease, reducing hyperoxaluria, preventing calcium oxalate stone formation, and improving kidney health in Grhpr knockout mice. N-Propargylglycine shows promise in research focused on neurodegenerative disorders, breast cancer, and metabolic syndromes.
