Anticancer Agent

NDI-Lyso is a lysosome-targeted anticancer agent that operates through an enzyme-instructed self-assembly (EISA) mechanism mediated by cathepsin B. This compound induces the formation of rigid long fibers within cancer cell lysosomes, leading to lysosomal swelling, membrane permeabilization, and disruption, ultimately resulting in apoptosis through a non-classical, caspase-independent pathway. NDI-Lyso demonstrates notable selective anticancer activity across various cancer cell lines, including drug-resistant variants, with an IC50 of approximately 10 μM, while exhibiting low toxicity toward normal cells (IC50 > 60 μM).
