U2AF-RNA Enhancer
NSC 194308 is an enhancer of U2AF2-RNA complexes that promotes the association of the U2AF1-U2AF2-SF1-splice site RNA complex by binding between the RNA recognition motifs of U2AF2. This compound impedes pre-mRNA splicing by stalling spliceosome assembly at the step where U2AF facilitates U2 snRNP recruitment to the branchpoint. NSC 194308 selectively induces cell death in leukemia cell lines harboring spliceosome mutations by enhancing the binding of pre-mRNA to U2AF2, making it a valuable tool for investigating spliceosomal regulation in cancer research.
