NV914
FTSJ1 Inhibitor
NV914 is a selective inhibitor of FTSJ1, a tryptophan tRNA-specific 2'-O-methyltransferase. By inhibiting the methyltransferase activity of FTSJ1, NV914 promotes translational readthrough of premature termination codons, allowing for the synthesis of full-length proteins in genes with nonsense mutations. This compound exhibits translational readthrough activity in vitro without affecting natural termination codons, thereby restoring CFTR protein expression. NV914 demonstrates favorable oral tolerance in murine models, indicating a low health risk and a good safety profile, making it valuable for research in cystic fibrosis and Shwachman-Diamond syndrome.
