Pan-RAS-IN-6 is a selective inhibitor of dual specificity phosphatase 6 (DUSP6), effectively reducing MAPK activation in the NCI-H1373-Luc model. This compound demonstrates significant tumor growth inhibition and regression in a non-small cell lung cancer (NSCLC) brain metastasis mouse model, highlighting its potential for therapeutic applications in KRAS mutation-driven cancers. With IC50 values of 1.3 nM, 4.7 nM, and 0.3 nM for KRAS G12C, G12D, and G12V mutants, respectively, Pan-RAS-IN-6 exhibits strong inhibitory effects on various KRAS mutation-related signaling pathways.
