BDK Inhibitor
PF-07208254 is a selective allosteric inhibitor targeting branched-chain ketoacid dehydrogenase kinase (BDK), with an IC50 of 110 nM. This compound enhances the catabolism of branched-chain amino acids (BCAAs) and branched-chain keto acids (BCKAs) by binding to BDK’s allosteric site, thereby inhibiting BDK-mediated BCKDH phosphorylation and promoting BDK degradation. PF-07208254 has demonstrated activity in human skeletal muscle cells (IC50 = 540 nM) and shows potential for improving cardiac function and metabolism. It is applicable for research in cardiometabolic diseases such as heart failure and type 2 diabetes.
