Phenformin

Antidiabetic/Antitumor Agent

Phenformin, a biguanide hypoglycemic agent, primarily targets mitochondrial respiratory chain complex I. This inhibition results in an increased AMP/ATP ratio, activating AMP-activated protein kinase (AMPK) and subsequently suppressing the mTOR pathway, which leads to reduced cell proliferation and promotes apoptosis and autophagy. Additionally, phenformin has shown effectiveness in inhibiting cancer stem cells (CSCs) and demonstrates promising antitumor activity, making it a valuable compound for research in metabolic disorders and cancer.
