Polθ-pol Inhibitor

Polθ-IN-10 is a selective inhibitor of Polθ-pol, demonstrating impressive potency with a human IC50 of 1.3 nM. By binding to the allosteric site of Polθ-pol, it disrupts the microhomology-mediated end-joining DNA repair pathway, further exhibiting inhibition of CYP2C9 (IC50=1.63 μM). Polθ-IN-10 effectively targets and hinders the proliferation of HR-deficient cancer cells, promoting apoptosis. This compound is valuable for research into the mechanisms and therapies surrounding HR-deficient cancers.
