PDK Inhibitor
PS10 is a potent ATP-competitive inhibitor of pyruvate dehydrogenase kinase (PDK), demonstrating effective inhibition across all PDK isoforms with IC50 values of 0.8 μM for PDK2, 0.76 μM for PDK4, 2.1 μM for PDK1, and 21.3 μM for PDK3. This compound exhibits a higher affinity for PDK2 (Kd = 239 nM) compared to Hsp90 (Kd = 47 μM). PS10 has been shown to enhance glucose tolerance and stimulate myocardial carbohydrate oxidation in models of diet-induced obesity, making it a valuable tool for researching diabetic cardiomyopathy.
