Rafutrombopag (tautomerism)

TPOR/MPL Agonist

Rafutrombopag is a nonpeptide agonist of the thrombopoietin receptor (TPOR/MPL) that exhibits oral bioactivity. This compound promotes hematopoiesis and alleviates iron overload by chelating iron, stimulating the proliferation and differentiation of TPOR-expressing cells, such as human hematopoietic stem cells. Rafutrombopag activates critical signaling pathways, including STAT, PI3K, and ERK, leading to the upregulation of cell cycle-related proteins and modulation of apoptosis. It also offers protective effects on cardiomyocytes under oxidative stress, making it a valuable tool for researching conditions such as immune thrombocytopenia and cardiovascular diseases linked to oxidative stress.
