Rafutrombopag
TPOR/MPL Agonist
Rafutrombopag is an orally active nonpeptide agonist of the thrombopoietin receptor (TPOR/MPL). This compound promotes hematopoiesis and alleviates iron overload by stimulating the proliferation and differentiation of human TPOR-expressing cells, including 32D-MPL and hematopoietic stem cells, through the activation of the STAT, PI3K, and ERK signaling pathways. Rafutrombopag also up-regulates key G1-phase proteins such as p-RB, Cyclin D1, and CDK4/6, facilitates cell cycle progression, and inhibits apoptosis by modulating BCL-XL/BAK expression. Additionally, Rafutrombopag enhances cardiomyocyte survival against oxidative stress, making it a significant tool for studies related to immune thrombocytopenia and oxidative stress-associated cardiovascular diseases.
