DKK3 LRP1 Inhibitor

SJ-300 is a selective inhibitor of the DKK3-LRP1 interaction, characterized by its oral activity and ability to penetrate the blood-brain barrier. This compound effectively restores amyloid-beta (Aβ) clearance in Alzheimer’s disease models while exhibiting a Kd of 7.9 μM for mLRPIV and an IC50 of 3.2 μM for inhibiting the DKK3-mLRPIV complex. Notably, SJ-300 does not interfere with Aβ binding to LRP1 and demonstrates significant cognitive improvement and reduction of neuropathology, including approximately 73.3% reduction in Aβ plaques in vivo. This reagent is poised for applications in Alzheimer’s disease research.
