ENL YEATS Inhibitor
TDI-11055 is a selective ENL YEATS inhibitor that effectively disrupts the interaction between the ENL protein's YEATS domain and acylated histones, leading to its displacement from chromatin. This compound exhibits IC50 values of 50 nM against the ENL YEATS domain and 70 nM against AF9, while demonstrating no activity towards GAS41 or YEATS2. TDI-11055 is valuable for studies on acute myeloid leukemia, as it reduces the chromatin occupancy of ENL-associated complexes, inhibits transcription elongation, and downregulates key oncogenic gene expression, promoting differentiation.
