cGAS Inhibitor
TDI-6570 is an orally active inhibitor of cyclic GMP-AMP synthase (cGAS), with a reported IC50 of 1.64 μM. This compound effectively penetrates the blood-brain barrier, demonstrating high gastrointestinal absorption and an extended half-life in murine models, while exhibiting no toxicity to primary neurons. By disrupting the cGAS-STING-IFN signaling pathway, TDI-6570 reduces STING expression and TBK1 activation, thereby inhibiting double-stranded DNA-induced cGAS activation and subsequent interferon-stimulated gene expression. This activity has implications for research into neurodegenerative and autoimmune disorders, including Alzheimer's disease and systemic lupus erythematosus, by potentially mitigating tau protein spread and enhancing cognitive function.
