Thioacetamide

Indirect hepatotoxin

Thioacetamide (TAA) is classified as an indirect hepatotoxin that primarily targets liver parenchymal cells, leading to necrosis. Its bioactivation occurs through the cytochrome P450 enzyme CYP2E1, converting TAA to reactive metabolites such as thioacetamide-S-oxide and thioacetamide-S-dioxide. These metabolites covalently interact with proteins and lipids, resulting in oxidative stress and centrilobular necrosis. Thioacetamide is widely used in research to model liver diseases, including chronic liver fibrosis and hepatic encephalopathy.
