Nuclear Pore Complex Dilator
trans-Cyclohexane-1,2-diol (TCHD) acts as a transient dilator of the nuclear pore complex (NPC) by targeting the hydrophobic core of FG nucleoporins. This activity disrupts NPC structure, leading to a reversible increase in nuclear pore permeability, which facilitates the passive diffusion of macromolecules exceeding 40 kDa, including plasmid DNA, into the nucleus. TCHD is particularly effective in enhancing the efficiency of in vitro electrotransfection and lipid-mediated gene transfection, offering significant improvement in gene expression within differentiated airway epithelial cells.
