TRV-120027

AT1R Antagonist

TRV-120027 is an AT1R antagonist that selectively engages β-arrestin-1 while inhibiting G-protein signaling pathways. This compound is known to induce acute catecholamine secretion through the activation of the TRPC3 cation channel, facilitating the formation of a macromolecular complex involving AT1R, β-arrestin-1, TRPC3, and PLCγ at the plasma membrane. TRV-120027 effectively inhibits angiotensin II-mediated vasoconstriction and enhances contractility in cardiomyocytes, highlighting its potential application for treating acute decompensated heart failure (ADHF).
