VraRC Inhibitor

TST1N-224 is a potent inhibitor of the response regulator VraRC, specifically disrupting VraRC-DNA complex formation with an IC50 of 60.2 μM. Through a fast-on-fast-off binding mechanism, TST1N-224 interferes with VraRC's binding to its cognate DNA, exhibiting a KD of 23.4 μM. It demonstrates significant antibacterial activity against Staphylococcus aureus, including methicillin-resistant strains (MRSA) and vancomycin-intermediate strains (VISA), with MIC values indicating effectiveness. Notably, TST1N-224 enhances the susceptibility of VISA to vancomycin and methicillin, positioning it as a valuable tool for research on antibiotic resistance mechanisms.
