VCP/p97 Inhibitor

VCP/p97 IN-3 is an allosteric inhibitor of the valosin-containing protein (VCP/p97), exhibiting potent inhibitory activity with IC50 values of 9 nM against wild-type VCP and 12 nM and 19 nM against mutant forms N660K and V474A/D649A, respectively. This compound enhances K48-linked ubiquitination and increases levels of cleaved caspase-3, thereby activating endoplasmic reticulum (ER) stress and the unfolded protein response (UPR). VCP/p97 IN-3 demonstrates antitumor efficacy by inhibiting growth in RPMI-8226 cell subcutaneous xenograft mouse models and is valuable for research into multiple myeloma.
