EBNA1 Inhibitor
VK-2019 is an orally bioavailable selective inhibitor targeting the Epstein-Barr virus nuclear antigen 1 (EBNA1). By disrupting the protein-DNA interface, VK-2019 interferes with the recruitment of viral DNA replication machinery, effectively blocking EBV replication in latently infected cells. This compound has been shown to reduce the copy number and gene expression of Epstein-Barr virus, decrease EBER-positive cells, and exhibit significant antiviral, immunomodulatory, and antiproliferative effects. In vivo studies demonstrate that VK-2019 inhibits tumor growth in EBV-dependent xenograft models, making it a valuable tool for research in advanced nasopharyngeal carcinoma and various EBV-associated malignancies.
