VTP50469 mesylate
Epigenetic Reader Domain Inhibitor
VTP50469 mesylate is a selective inhibitor of the Menin-MLL1 interaction, primarily targeting MLL-rearranged and NPM1c+ leukemias. This compound effectively induces cell death in cell lines harboring MLL rearrangements and NPM1c+ mutations by displacing Menin from protein complexes and inhibiting MLL's chromatin occupancy at specific gene loci. The resultant alterations in gene expression lead to enhanced differentiation and apoptosis. In preclinical studies using patient-derived xenograft models, VTP50469 has demonstrated substantial reductions in leukemia burden and prolonged disease-free survival in treated subjects.
