WRN Inhibitor

WRN-IN-24 is an orally active allosteric covalent inhibitor of Werner syndrome helicase (WRN), exhibiting an IC50 of 201 nM. By binding to a unique allosteric pocket and forming a hydrogen bond with K894, WRN-IN-24 effectively inhibits WRN activity. This compound demonstrates significant antiproliferative effects on colorectal cancer cells and shows dose-dependent antitumor efficacy in xenograft mouse models. WRN-IN-24 is a valuable tool for investigating microsatellite instability-high cancers, particularly in colorectal cancer research.
