DESI2 Inhibitor
WWQ-03-012 is a selective inhibitor of the deSUMOylating isopeptidase DESI2, exhibiting an IC50 of 47.3 μM. This compound effectively induces ubiquitination and proteasomal degradation of the JAK2-V617F mutation while sparing wild-type JAK2, leading to the blockade of JAK2-STAT3/5 signaling pathways. WWQ-03-012 also demonstrates antiproliferative effects and promotes apoptosis in cancer cells. It shows synergistic activity when combined with Ruxolitinib, enhancing its cytotoxicity against JAK2 mutant cells, making it a valuable tool for cancer research, particularly in myeloproliferative neoplasms.
