XAN-5

MtG4 Ligand

XAN-5 is a selective mitochondrial DNA G-quadruplex (mtG4) ligand with a dissociation constant (Kd) of 3.8 μM that binds to and stabilizes mtG4 structures. This compound disrupts mitochondrial gene transcription and DNA replication, leading to mitochondrial dysfunction, increased reactive oxygen species (ROS) production, cell cycle arrest in the G0 phase, and caspase-dependent apoptosis. Additionally, XAN-5 inhibits autophagy and promotes immunogenic cell death while significantly reducing tumor growth in a murine model of liver cancer, enhancing infiltration of CD4+ and CD8+ T cells. XAN-5 serves as a valuable tool for investigating therapeutic strategies in liver cancer research.
