XL-3158
cGAS Inhibitor
XL-3158 is a selective cyclic GMP-AMP synthase (cGAS) inhibitor, demonstrating an IC50 of 11.1 μM for human cGAS and 2.19 μM for mouse cGAS. It functions by occupying both allosteric and orthosteric sites on cGAS, stabilizing its activation loop in an inactive conformation, which disrupts cGAS-DNA interactions and targets phase separation. XL-3158 effectively reduces the local concentration of cGAS within cells by inhibiting aggregate formation. This reagent is valuable for investigating cGAS-dependent inflammatory diseases.
