RANKL Inhibitor
Y1693 is a selective inhibitor of RANKL, exhibiting a Kd of 5.03 μM for human RANKL. It effectively disrupts the c-fos/NFATc1 signaling pathway by blocking its interaction with RANK, leading to reduced osteoclast differentiation and bone resorption. Y1693 downregulates mRNA and protein levels of TRAP, cathepsin K, c-fos, and NFATc1 without inducing significant cytotoxicity in bone marrow-derived macrophages and osteoclast precursors. This compound demonstrates therapeutic potential in osteoporosis and periodontal disease research, as evidenced by its efficacy in reversing ovariectomy-induced osteoporosis and ligation-induced periodontal bone loss in murine models.
